L-Glutamin-Induced Inhibition of Endotoxemia by Reducing Pre-Inflammatory Cytokines, Nitric Oxide, and CD40 Expression

Authors

Abstract:

Introduction: Researchers have been investigating the major pathogens involved in septic shock caused by lipopolysaccharide in recent years. Moreover, they have been concerned about the use of appropriate drugs to intervene with the main pathogenesis of the disease. This study aimed to investigate the effect of L-Glutamine as an HSP70 inducer in reducing pre-inflammatory cytokines and effective intermediaries in septic shock, including nitric oxide production and CD40 expression in mice treated with lipopolysaccharide (LPS).   Materials & Methods: In total, 25 Swiss Albino mice were randomly divided into three groups of A (control group, n=5), (received LPS with a dose of 0.75 mg/animal intraperitoneally [IP], (n=10), and (received L-glutamine with a dose of 50 mg/kg IP 30 minutes after the injection of LPS [three doses every 24 h], (n=10). Subsequently, one day after the final dose of drug induction, the number of cytokines, such as IFN-γ, IL-4, IL-10, HSP70, CD40, and nitric oxide, were evaluated in this study. Ethics code: IR.TABRIZU.REC.1398.003   Findings: The data were analyzed in SPSS software (version 19) through a one-way analysis of variance and Scheffe test. According to the results, L-glutamine can significantly reduce the serum levels of pre-inflammatory cytokine, nitric oxide, and CD40 expression in the drug-treated group (group C), compared to the LPS- treated group (group B) (P<0.05).   Discussions & Conclusions: L-glutamine could be a good choice for treatment by reducing the level of inflammatory mediators.

Inhibition of Nitric Oxide Production and Proinflammatory Cytokines by Several Medicinal Plants

Background: A number of medicinal plants have been used to treat various immunological diseases. Nitric oxide (NO) has an important regulatory role in the various types of inflammatory processes. Objective: To investigate the NO modulatory activity of the extracts of several medicinal plants native to Iran including Dracocephalum kotschyi, Linum persicum, Dionysia termeana, Salvia mirzayanii, F...

Inhibition of pro-inflammatory cytokines and inducible nitric oxide by extract of Emilia sonchifolia L. aerial parts.

Emilia sonchifolia L. (Asteraceae) is used in ethnomedicine for the treatment of a wide array of inflammatory disorders. This practice has also been supported by scientific reports which showed that extracts of E. sonchifolia possess anti-inflammatory effects in rodents. However, the mechanism(s) through which the extracts produce these effects is not known. In this study, the effect of a metha...

Nitric oxide measurements during endotoxemia.

BACKGROUND Excessive continuous NO release from inducible NO synthase over prolonged periods under pathological conditions, such as endotoxemia, contributes significantly to circulatory failure, hypotension, and septic shock. This NO production during endotoxemia is accompanied by superoxide release, which contributes to the fast decay of NO. Therefore, the amount of NO that diffuses to target ...

Expression of inducible nitric oxide synthase and inflammatory cytokines in alveolar macrophages of ARDS following sepsis.

STUDY OBJECTIVE The objective of this study was to evaluate the role of inducible nitric oxide synthase (iNOS) and proinflammatory cytokines in alveolar macrophages (AMs) in the pathogenesis of ARDS following sepsis. SETTING ICU in a university hospital. DESIGN Prospective exploratory, open-labeled study was carried out. PATIENTS A total of 24 patients were investigated: 8 patients diagno...

Nitric oxide decreases cytokine-induced endothelial activation. Nitric oxide selectively reduces endothelial expression of adhesion molecules and proinflammatory cytokines.

To test the hypothesis that nitric oxide (NO) limits endothelial activation, we treated cytokine-stimulated human saphenous vein endothelial cells with several NO donors and assessed their effects on the inducible expression of vascular cell adhesion molecule-1 (VCAM-1). In a concentration-dependent manner, NO inhibited interleukin (IL)-1 alpha-stimulated VCAM-1 expression by 35-55% as determin...
